Preparation | N-acetic acid -3-indole formaldehyde can be prepared by the nucleophilic substitution reaction of 3-formyl indole and ethyl bromoacetate (3-formyl indole -1-yl) ethyl acetate, and then hydrolyze the ester group to prepare N-acetic acid -3-indole formaldehyde. 3-formyl indole (10g,69mmol) was dissolved in N,N-dimethylformamide (100mL), and the temperature was kept below 15 ℃ under external cooling under nitriding atmosphere. Sodium hydride (60%,3.0g,76mmol in mineral oil) was added in batches. Then, after 30 minutes, ethyl bromoacetate (8.4mL,76mmol) of N,N-dimethylformamide (15mL) solution was added dropwise, and the resulting mixture was stirred at room temperature for 16 hours. Concentrate the mixture in vacuum so that the residue is divided between water (300mL) and ethyl acetate (2 × 150mL). The organic extract was combined, washed with saturated ammonium chloride aqueous solution (100mL), dried (MgSO4), and concentrated in vacuum to obtain 15.9g (quantitative) (3-formylindole-1-yl) ethyl acetate, an oil. 1H-NMR(CDCl3):δH = 1.30(3H,t),4.23(2H,q),4.90(2H,s),7.3(3H,m),7.77(1H,s),8.32(1H,d),10.0(1H,s). will (3-formyl indole -1-yl) ethyl acetate (15.9g,69mmol) is dissolved in 1,4-dioxane (100mL), 1N sodium hydroxide (10mL) is added, and the obtained mixture is stirred at room temperature for 4 days. Water (500mL) was added and the mixture was washed with diethyl ether (150mL). The aqueous phase was acidified with 5N hydrochloric acid and extracted with ethyl acetate (250 150mL). The organic extract was combined, dried (MgSO4) and concentrated in vacuum to obtain 10.3g(73%) N-acetic acid -3-indole formaldehyde as solid. 1H-NMR(DMSO-d6):δH=5.20(2H,s),7.3(2H,m),7.55(1H,d),8.12(1H,d),8.30(1H,s),9.95(1H,s),13.3(1H,bs). |
